چکیده

Aminoglycosides are a class of antibiotics that inhibit protein synthesis in bacteria by binding to the 30S ribosomal subunit. However, it has been reported that certain aminoglycosides have secondary effects on the cell’s overall carbohydrate concentration. Previous studies have also reported an increase in capsule size. This study attempted to investigate the effects of kanamycin and streptomycin on carbohydrate production, particularly the sugars found in the capsular polysaccharides. Escherichia coli B23 was incubated in the presence of kanamycin and streptomycin. It was found that the concentration of glucuronic acid in the capsule increased in both the kanamycin and streptomycin treated cells, which correlated with the increase in the concentration of carbohydrate in the capsule and in the cell. However, a decrease in the amount of capsular 6-deoxyhexose was observed in streptomycin treated cells, correlating with a decrease in the overall concentration of 6-deoxyhexose. _______________________________________________________________

The Effect of Sub-Inhibitory Streptomycin on Capsular Polysaccharide Production and Streptomycin Resistance in Escherichia coli

Previous studies have shown that pre-treatment of Escherichia coli B23 with subinhibitory concentrations of aminoglycosides leads to a subsequent increase in the level of capsular polysaccharide, as well as an increased resistance to streptomycin. Using capsular polysaccharide synthesis pathway mutants, we investigated the correlation between the level of capsular polysaccharide and streptomyci...

The Effect of Aberrant Protein Synthesis on the Production of Capsular Polysaccharide in Escherichia coli B23

Streptomycin, an aminoglycoside, binds to and modifies prokaryotic ribosomes leading to a decrease in protein synthesis and the production of mistranslated proteins. Previous studies reported that Escherichia coli B23, under sub-lethal concentrations of streptomycin, exhibited an increase in capsule polysaccharide (CPS). The mechanism through which this increase in CPS occurs was unknown. In th...

Sublethal Kanamycin Induced Cross Resistance to Functionally and Structurally Unrelated Antibiotics

Previous studies have shown that pretreatment with sub-lethal kanamycin to E. coli B23 cells provided protection against subsequent exposure to lethal amounts of the same antibiotic. It has also been shown that exposure to sub-inhibitory levels of certain antibiotic can induce cross resistance by promoting the development of low-level resistance to other functional and structural unrelated anti...

Persistence of Antibiotic Resistance and Capsule in E. coli B23 after Removal from Sublethal Kanamycin Treatment

Kanamycin belongs to the aminoglycoside class of bacteriostatic antibiotics that inhibits protein synthesis. Previous studies have shown that treatment with sublethal levels of kanamycin causes the extracellular polysaccharide capsule in E. coli B23 to increase in size, and potentially confers the bacterium with kanamycin resistance. This study attempted to confirm this by establishing that inc...

Expression of E.coli capsular polysaccharide requires the KfiB protein:A Structural based analysis

Abstract Background and objectives: important virulence factor for many invasive bacterial pathogens of humans. Escherichia coli offer a model system to study the mechanisms by which capsular polysaccharides are synthesized and exported onto the cell surface of bacteria. Biosynthesis of the E consists of the repeat structure -4) GlcA- (1, 4)-GlcNAc- (1-, requires the KfiA,...
